The Serra do Divisor National Park is situated in western Brazil's Amazon plain, close to the border with Peru. It forms one of the Amazon's most extensive and unbroken protected areas when combined with Peru's Serra del Divisor National Park.

Serra do Divisor National Park is located in the Amazon Plain, near the border with Peru. Established in 1989, it covers an area of 843,012 ha (2,083,128 acres) in the state of Acre, at the westernmost point of Brazil. It also contains the highest point in the state of Acre, reaching 609 m (1,998 ft) above sea level.

The National Park is situated within the Sierra del Divisor. This mountain range runs along the border between Peru and Brazil that rises from the plains of the Amazon drainage basin.

Serra do Divisor National Park adjoins Serra del Divisor National Park, located across the border in eastern Peru. Combined, the two National Parks make up one of the largest contiguous blocks of protected areas in the Amazon.

The National Park adjoins the Alto Juruá Extractive Reserve along its southeast border. The Juruá River defines the eastern boundary of the Park's southern section, and the Azul River defines the eastern boundary of the northern area.

The primary access to the National Park is by boat from the City of Cruzeiro do Sul via the Môa River or Juruá River. There is no tourism infrastructure.

Serra do Divisor National Park is in the Southwest Amazon moist forests ecoregion. Most of the area is covered by open rainforests with palm trees or bamboo, dense and open sub-mountain rainforests, and dense and open alluvial rainforests (periodically-inundated forests).

The area experiences a hot and humid tropical climate, with one or two months of dry season. Average annual temperatures are more than 24 °C (75 °F). Annual rainfall is between 1,750 - 2,000 mm (69 - 79 in).

An indigenous population of approximately 1,200 live within or at the margins of the National Park.
